BIOINFORMATICS PERSPECTIVE ON THE RELATIONSHIP BETWEEN GVHD AND CXCL8 (IL-8)

Graft-versus-host disease is a phenomenon that develops after stem cell transplantation. In allogeneic transplants, where a person receives a transplant from someone else, it occurs when active T lymphocytes from the donor recognise the recipient’s tissues as foreign and attack them. GENEMANIA database was used to determine the relationship of IL-8 cytokine with other genes, STRING database to understand its relationship with other genes (pathways used, physical interactions, gene ontology, etc.) and GeneCard databases and GWAS/GRAPS to show its role in diseases. In addition MalaCard and DisGeNET were used to find genes associated with GvHD. UniProt and KEGG were used for the association with pathways. MalaCard and DisGeNET showed that GvHD is associated with molecules such as IL-1A, IL-4, IL-6, IL-8, IL-9, IL-10, IL-17A, IL-12, IL-18, AHR, ITGA, DCD19, CD83, CD274, TLR6, TLR9, STAT3, FOXP3, REG3a, NLRP, HSP90, VCAM1, ITGB1, HIF1a. CXCL8, also known as IL-8, has been shown in STRING and GeneCard Database to communicate strongly with IL-1, IL-6, IL-10, TNF and CXCL 8 receptors CXCR1 and CXCR2. STRING Database, which processed information from KEGG Pathway and Uniport, found that CXCL 8 was significantly associated with Chemotaxis (FDR=6.68e-05), Cytokine (FDR=2.47e-07), Inflammatory response (FDR=0.0156), Interleukin-1 processing (FDR=0.0020), CD163 mediating an anti-inflammatory response (FDR=0.0023), Interleukin-10 signaling (FDR=2.29e-10). This gene is overexpressed in Whole Blood (x26.0). Also IL-8 is expressed in Leukocyte (FDR=0.0345), Haematopoietic system (FDR=2.16e-06), Bone marrow-derived macrophage (FDR=0.00028). IL-8 has been shown to be closely associated with melanoma. Cytokines are highly associated with haematological malignancies, especially with GvHD, a transplant-related disease after stem cell transplantation. In fact, recent studies support this. Experimental study of the relationship of cytokines with other molecules and pathways in the laboratory environment will guide the pathogenesis and treatment of GvHD.
